Diving Tulamben or Padang Bai? No (for most people).

Nusa Penida? Lembongan? Yes (for most people).
 
Hi, Absolutly ! At least a 5 mm
East coast ( Padang bai and Candidasa ) Nusa Lembongan & Penida the water temperature on those dates can drop till 22ºC with unpredictable termoclines under 17ºC .
North east coast ( Tulamben and Amed ) Is more stable, normal temparature of the water all year around 26ºC to 28ºC but I have to admit that sometimes even there I haven been surprise for lovely cold termoclines :)
North-West ( Menjangan ) Always warm 28-30ºC

I have noted that you would like to know about the water temperature in Bali on mid of October.

For Tulamben Bay and Tulamben area, the water temperature will be approx. 25 - 30 degree Celsius so the 3mm wetsuit will be good enough for you. However, for Nusa Penida and Nusa Lembongan area, the water temperature will be approx. 20 - 25 degree Celsius therefore I would suggest you to bring the 5mm wetsuit for diving on this area.

I am a huge fan of traveling to Bali with a 3 mil and a 2.5 mil hooded vest. The hooded vest is perhaps the greatest invention ever as it weighs almost nothing and takes far less room than a 3 mil and a 5 mil.
 
Apologies for resuscitating this post.

I will be heading up to Nusa Penida in August. It will be my first dive at Nusa Penida. I was advised to get a hood and/or a 5mm suit to prepare for Nusa Penida. Would that be necessary?
I'm thinking of going with a Thermal Layer + 3mm suit + Gloves. My suit is fairly snug so water usually can't slosh in unless I allow it to.

Previously I have dived at Tulamben and Amed with just a thermal layer and board-shorts and at times it got chilly but most of the time its fine.

August in Nusa Penida can often get to 19C or less.. coldest I have had at Crystal Bay is 16C, coldest of someone else I know is 13C. The sites in and around the channel are typically the coldest. Be prepared! If you don't want to bring your own 5mm then ask your dive operator if they have one you can rent
