I'm planning on a Manta Ray Night dive, in Hawaii, with my GoPro Hero 4 Black.
1. What will be the best setting?
2. They supply a 1,200 Lumen strobe, is that enough or do I need more light?

Hiya Chocolady. I do this manta night dive/snorkel once a week here in Kona. You will be fine with the light given...as long as you are ok doing video instead of stills. Video really is the way to go if you are just going to do this dive once. It works much better in low light with fast moving subjects, especially ones that are black on one side and white on the other. I've gone down with an canon 5d mark ii with two strobes and its still a tough place to get still pictures. Here is a clip I shot with my GoPro hero 4 silver: You will have a blast. So take high resolution video and sit back and enjoy the show.
