Under left arm, underneath the harness. It stays there no matter what happens, on or off...
 
Probably doesn't matter as much for singles (I do it from the left there as well), but when wearing doubles, I want it coming off the opposite first stage as my wing inflator, so I have redundant buoyancy. My wing inflator is from the right post, so my drysuit inflator comes from the left post (and under the left arm).
 
Under my left arm. I'm right handed so when I shoot a bag, I hold the spool and bag in my right hand, unhook the DS inflator with my left then put air in the bag. I can then connect it back with my left hand while holding the spool in my right. That's why it's routed under my left arm and under my left chest strap.
