PTFE thread sealing tape on his?

Do I need to do anything to this setup other then put it together and make sure its "02" clean? Do I need to add the tape or? This is my backup for 02 emergencies or transfilling.

PTFE tape is not for sealing, it is for friction control when tightening compression threads for NPT.
You will need it between the brass CGA adapter and what I assume is the DIN outlet but not the CGA-540 adapter.
PTFE tape does not seal? Please do explain.

the actual O2 clean stuff is the same tape but handled differently. If I was building a unit for commercial use in a shop then I would certainly use it. Personally I bought some for gas connections, but if I'm in a pinch then I'll use regular stuff

 
The O2 tape I have is noticably thicker than the stuff from the local home center. Has a slightly different feel to it as well. I got this before the covid price adjustment
For reference I have had an accidental Oxygen fire once. I try to avoid repeats. It wasn't teflon, it was an O-ring that lit up. But still I now take a little more caution after the incident.

The thin cheap stuff is a real pain. The the thick gas tape usually yellow is way to thick...
there is nice stuff that that's in between that I like, but can be hard to find.
My hydraulic guys have stuff that quite good.
But not as good as a bunch I got from a truck shop, it was excellent stuff but think it was old stock..
